Plant tissue culture is based on the property of the plant cells to regenerate a whole plant which is known as totipotency

To initiate tissue culture for asexual reproduction, single cells, plants cells without cell walls (protoplasts), pieces of leaves, small parts of stems or roots are taken to generate a new complete plant on the culture media in the presence of nutrients and plant hormones.
