The steps of preparation of temporary mount of a leaf peel to observe stomata are as follows:- 1. Take a healthy leaf from your garden or the potted plant. 2. Fold the leaf and remove a part of the peel from the lower surface of the leaf using forceps. 3. Leave the peel for some time in a watch glass containing water. 4. Add a few drops of safranin stain in the sample through a dropper. 5. After 2-3 minutes, take the peel out and set it on a clean glass slide. 6. Add a drop of glycerin over the peel, and put a clean coverslip gently over it using a needle. 7. Remove the excess glycerin and stain using blotting paper. 8. Observe the slide first under a low-power, and then under a high-power magnification of a compound microscope.
